Abstract

The aim of this work was to investigate the effects of different concentrations of sucrose, combined with different photoperiods, on in vitro sprouting rate of “MM.111” apple rootstock. To set up the experiment, sproutings of about 1.5 cm in length were inoculated in test tubes containing 10 ml of “MS” medium and supplied with five concentrations of sucrose (0.0, 15, 30, 45 and 60 g/L). Afterwards, these tubes were allocated to B.O.D. chambers with photoperiods adjusted to 8/16, 10/14, 14/10 and 16/8 light/dark hours, at 25ºC temperature and at light intensity of 1500 lux. The experimental design was completely randomized in a 5×4 factorial scheme. At the end of a 30-day period, the experiment was evaluated by registering the total number of sprouts and the number of sprouts ≥ 1.0 cm in length. The results were interpreted through a variance and regression statistical analysis. The best combination for the total number of sprouts and the number of sprouts ≥ 1,0 cm in length for the “MM.111” apple rootstock was 60 g/L with an 8-hour photoperiod.
